Staphylococcal variable number tandem repeat (VNTR)-spa genotyping and their role in phylogenic study

Staphylococcus aureus protein A is considered a vital virulence element determinant of its pathogenicity. Its sequence diversity aids in staphylococcal typing and phylogeny. The present study aimed to the genotyping method for S. isolates by applying Spa (variable number tandem repeat) their role phylogenic study. Twenty were achieved from various clinical subjected complete identification diagnosis. Later on, these DNA extraction PCR amplification sequencing SPA gene. showed that out 20 amplified spa gene, 8 different types among 18/20 detected 2/20 could not be typed, as most commonly observed t304 (35%), t491 (15%) followed t078 t059 (10%). Finally, depending on bacterial phylogenetic relationships, categorized into 2 clades. first one contained 18 second isolates. Most included clade (18 isolates) whereas only involved B. grouped 3 groups established dissimilarity repeats Cluster 1 t304, t078, t044 types, cluster t059, t4870 t386 t091 types.
